Immunogen
This GLI1 antibody was raised against a peptide of human GLI1.
Buffer
Purified IgG in 140 mM Hepes, pH 7.5, 70 mM NaCl, 32 mM NaOAc, 40% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.
AbFlex® GLI1 rAb antibody tested by Western blot.
40 µg of SJSA-1 whole cell extracts were probed with AbFlex® GLI1 antibody at 0.5 µg/mL.
AbFlex® GLI1 rAb antibody tested by immunoprecipitation.
2 µg of antibody was used to immunoprecipitate GLI1 from 1 mg of SASJ-1 whole cell extract, 40% IP product (Lane 2) was then run on SDS-PAGE and probed with the same antibody at 0.5 µg /ml. Tween-20 at 0.1% was added to blocking buffers to perform Western blot and reduce background. 2 µg of rabbit IgG was used as negative control for IP (Lane 2).
AbFlex® GLI1 rAb antibody tested by immunofluorescence.
GLI1 was detected in human HeLa cells using AbFlex® GLI1 antibody at 2 µg/mL. Followed by Multi-rAb CoraLite® Plus 594-Goat Anti-Rabbit Recombinant Secondary Antibody (H+L).DAPI at 594 nm.
AbFlex® GLI1 rAb antibody tested by ChIP-Seq.
ChIP was performed using ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 18 µg of chromatin from SJSA-1 cells and 2 µg of GLI1 antibody. ChIP DNA was sequenced on the Illumina NovaSeq and 43 million sequence tags were mapped to identify GLI1 binding sites. The image shows binding across a region of chromosome 1.
Background
AbFlex® antibodies are recombinant antibodies (rAbs) that have been generated using defined DNA sequences to produce highly specific, reproducible antibodies. Each AbFlex® antibody contains a 6xHis Tag, a Biotinylation Tag forenzymatic biotin conjugation using the biotin ligase, BirA, and a sortase recognition motif (LPXTG) to attach a variety of labels directly to the antibody including fluorophores, enzymatic substrates (HRP, AP), peptides, drugs as well as solid supports.
AbFlex® GLI1 antibody was expressed as full-length IgG with rabbit immunoglobulin heavy and light chains in mammalian HEK293 cells.
GLI1 (GLI family zinc finger 1, also known as PPD1, PAPA8) is a member of the Kruppel family of zinc finger proteins. GLI1 is activated by the sonic hedgehog signal transduction cascade. It plays a role in craniofacial development and digital development, as well as development of the central nervous system and gastrointestinal tract. The activity and nuclear localization of this protein is negatively regulated by p53 in an inhibitory loop.
