Immunogen

This MAX antibody was raised against a peptide of human MAX.

Buffer

Purified IgG in 140 mM Hepes, pH 7.5, 70 mM NaCl, 32 mM NaOAc, 40% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

 

AbFlex® MAX rAb antibody tested by Western blot.
40 µg of PC-3 whole cell extracts were probed with AbFlex® MAX antibody at 0.5 µg/mL.

AbFlex® MAX rAb antibody tested by immunofluorescence.
MAX was detected in human HeLa cells using AbFlex MAX (rAb) at 2 µg/mL, followed by Multi-rAb CoraLite® Plus 594-Goat AntiRabbit Recombinant Secondary Antibody (H+L).

AbFlex® MAX rAb antibody tested by ChIP-Seq.
ChIP was performed using ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 18 µg of chromatin from HEK293 cells and 2 µg of MAX antibody. ChIP DNA was sequenced on the Illumina NovaSeq and 21 million sequence tags were mapped to identify MAX binding sites. The image shows binding across a region of chromosome 12.

Background

AbFlex® antibodies are recombinant antibodies (rAbs) that have been generated using defined DNA sequences to produce highly specific, reproducible antibodies. Each AbFlex antibody contains a 6xHis Tag, a BiotinylationTag forenzymatic biotin conjugation using the biotin ligase, BirA, and a sortase recognition motif (LPXTG) to attach a variety of labels directly to the antibody including fluorophores, enzymatic substrates (HRP, AP), peptides, drugs as well as solid supports.

AbFlex® MAX antibody was expressed as full-length IgG with rabbit immunoglobulin heavy and light chains in mammalian HEK293 cells.

MAX (MYC Associated Factor X, also known as BHLHd4) is a member of the basic helix-loop-helix leucine zipper (bHLHZ) family of transcription factors. It is able to form homodimers and heterodimers with other family members, which include Mad, Mxi1 and Myc. MAX forms a sequence-specific DNA-binding protein complex with MYC or MAD which recognizes the core sequence 5-CAC[GA]TG-3. The MYC:MAX complex is a transcriptional activator, whereas the MAD:MAX complex is a repressor. May repress transcription via the recruitment of a chromatin remodeling complex containing H3 Lys-9 histone methyltransferase activity.

Storage

Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20°C for up to 2 years. Keep all reagents on ice when not in storage.
