Immunogen

This SIN3A antibody was raised against a peptide derived from the N-terminus of human SIN3A.

Buffer

Rabbit serum containing 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

Chromatin immunoprecipitation (ChIP) was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 30 µg of chromatin from TM4 Sertoli cells and 5 µl of SIN3A antibody. ChIP DNA was used in qPCR with the control primer pairs or gene-specific primer pairs as indicated. Data are presented as Binding Events Detected per 1000 Cells using Active Motif's Epigenetic Services normalization scheme which accounts for primer efficiency and the amount of chromatin used in the ChIP reaction.

Background

SIN3A (Switch independent 3 homologue) is the mammalian homologue of the yeast protein Sin3. SIN3A is transcriptional co-repressor that is recruited to promoters by sequence-specific DNA binding proteins, such as REST, MXI1 and MAD-MAX. It is part of a large multi-subunit repressor protein complex that also contains HDAC1, HDAC2 and SMRT/NCoR1.

Positive Control

Active Motif's HeLa nuclear extract (Catalog No. 36010) can be used as a positive control for SIN3A antibody.

qPCR Primers

Active Motif's Human Negative Control Primer Set 1 (Cat. No. 71001) has been validated for performing qPCR and endpoint PCR when this antibody has been used for ChIP on human samples. For ChIP experiments with this antibody on mouse samples, the Mouse Negative Control Primer Set 1 (Cat. No. 71011) has been validated for both qPCR and endpoint PCR.
