Background
The recombinant YAP1 / TEAD1 complex functions as a key transcriptional effector of the Hippo signaling pathway, where YAP1 acts as a co-activator that binds TEAD1 (its DNA-binding partner) to regulate genes controlling cell proliferation, organ size, and tissue regeneration. This complex drives repair in organs like liver and bone by activating stem cells and suppressing senescence, while its dysregulation promotes cancer progression. Its dual roles make it both a therapeutic target for oncology (via inhibition) and a tool for regenerative medicine.

Protein Details
Recombinant YAP1 / TEAD1 Complex that includes full length human YAP1 protein (accession number NP_001181973.1) with an N-terminal FLAG tag and full length human TEAD1 protein (accession number NP_068780.2) with an N-terminal His tag was expressed in Hi5 cells. The molecular weights of YAP1 and TEAD1 are 54.04 kDa and 50.74 kDa, respectively.

1 µM substrate dsDNA (5’ biotin GTCCACATTCCACCGAAG 3’) was incubated with different concentrations of YAP1 / TEAD1 Complex in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 0.1% BSA at room temperature for 1 hour, Anti-FLAG antibody was used to detect reaction products.
