Background
The recombinant YAP1 / TEAD2 complex functions as a transcriptional regulator critical for development and tissue homeostasis. As part of the Hippo signaling pathway, YAP1 (a transcriptional co-activator) binds TEAD2 (a DNA-binding transcription factor) to drive expression of target genes governing cell proliferation, stem cell maintenance, and organ size control. This complex plays specialized roles in embryonic development, neural differentiation, and muscle regeneration by activating programs for cell survival and plasticity. Dysregulation contributes to tumorigenesis in multiple cancers through hyperactivation of growth-promoting genes. Recombinant YAP1/TEAD2 is leveraged to study Hippo pathway mechanisms, model regenerative processes (e.g., neural/muscle repair), and screen therapeutic modulators for cancer or regenerative medicine applications.

Protein Details
Recombinant YAP1 / TEAD2 Complex that includes full length human YAP1 protein (accession number NP_001181973.1) with an N-terminal FLAG tag and full length human TEAD2 protein (accession number NP_001243587.1) with an N-terminal His tag was expressed in Hi5 cells. The molecular weights of YAP1 and TEAD2 are 54.04 kDa and 52.8 kDa, respectively.

1 µM substrate dsDNA (5’ biotin GTCCACATTCCACCGAAG 3’) was incubated with different concentrations of YAP1 / TEAD2 Complex in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 0.1% BSA at room temperature for 1 hour, Anti-FLAG antibody was used to detect reaction products.
