Background
The recombinant YAP1 / TEAD4 complex functions as a transcriptional regulator central to the Hippo signaling pathway, orchestrating cell proliferation, tissue regeneration, and organ development. YAP1, a transcriptional co-activator, partners with TEAD4 (a DNA-binding transcription factor) to drive expression of target genes governing stem cell maintenance, tissue repair (e.g., liver regeneration via hepatocyte-to-ductal reprogramming and bone regeneration through osteogenic gene activation), and embryonic development (e.g., trophectoderm specification via Tead motif binding). Dysregulation of this complex promotes pathological processes: hyperactivation drives cancer progression (e.g., triple-negative breast cancer migration and glioblastoma invasion) by upregulating oncogenes like TRIO to activate Rho GTPase-mediated cell migration and metastasis. The recombinant complex is utilized to study Hippo signaling mechanisms, model regenerative therapies (e.g., gene delivery for osteoarthritis), and screen inhibitors targeting YAPTEAD interactions in cancers. Its dual roles underscore therapeutic potential in regenerative medicine and oncology.

Protein Details
Recombinant YAP1 / TEAD4 Complex that includes full length human YAP1 protein (accession number NP_001181973.1) with an N-terminal FLAG tag and full length human TEAD4 protein (accession number NP_003204.2) with an N-terminal His tag was expressed in Hi5 cells. The molecular weights of YAP1 and TEAD4 are 54.04 kDa and 51.67 kDa, respectively.

HTRF Assay for Recombinant YAP1 / TEAD4 Complex activity. 1 µM substrate dsDNA (5’ biotin GTCCACATTCCACCGAAG 3’) was incubated with different concentrations of YAP1 / TEAD4 Complex in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 0.1% BSA at room temperature for 1 hour, Anti-FLAG antibody was used to detect reaction products.
