Background

DHX15 (ATP-dependent RNA helicase DHX15) is a multifunctional DEAH-box RNA helicase involved in several critical steps of RNA metabolism. It plays an essential role in pre-mRNA splicing as a core component of the spliceosome, where it is responsible for catalyzing the release of the lariat intron after splicing catalysis, a key step in spliceosome disassembly. Beyond splicing, DHX15 is a crucial cytoplasmic sensor for viral RNA, initiating innate immune defense pathways by activating NF-κB and MAPK signaling to induce pro-inflammatory cytokines and type I interferons. Its additional functions in ribosomal RNA processing and nonsense-mediated mRNA decay further highlight its importance in maintaining mRNA fidelity and overall cellular homeostasis.
