Background

RIGI (Antiviral innate immune response receptor RIG-I) is a crucial cytosolic pattern recognition receptor (PRR) that acts as a primary sensor of viral RNA in the innate immune system. It specifically recognizes short double stranded RNA (dsRNA) and single-stranded RNA with 5’-triphosphate (5’-ppp RNA) ends, which are characteristic of many viral genomes. Upon ligand binding, RIG-I undergoes a conformational change and activates the mitochondrial antiviral-signaling (MAVS) protein, triggering a downstream signaling cascade that leads to the robust production of type I interferons (IFN-α/β) and pro-inflammatory cytokines. This response is essential for establishing an antiviral state in infected and neighboring cells. Dysregulation of RIG-I signaling is associated with autoimmune disorders, chronic inflammation, and inadequate responses to viral infections.
