Background
TEAD3 (Transcriptional enhancer factor TEF-5) is a transcription factor central to the Hippo signaling pathway, which regulates organ size control, cell proliferation, apoptosis, and tissue regeneration. As a member of the TEAD family, it functions as the primary nuclear receptor for transcriptional co-activators YAP and TAZ. When the Hippo pathway is inactive, TEAD3 binds YAP/TAZ to form a complex that activates genes promoting cell growth, migration, and survival. It specifically regulates developmental processes, including trophoblast differentiation in the placenta and neural crest formation. Dysregulation of TEAD3 drives tumorigenesis in cancers such as mesothelioma, non-small cell lung cancer, and ovarian cancer by enabling uncontrolled proliferation and metastasis. Its conserved hydrophobic pocket is a key target for inhibitors against Hippo pathway-driven cancers.

Protein Details
Recombinant TEAD3 protein was expressed in a baculovirus expression system as the full length protein (accession number NP_003205.2) with an N-terminal FLAG tag. The molecular weight of TEAD3 is 50.06 kDa.

HTRF Assay for Recombinant TEAD3 protein activity
1 µM substrate dsDNA (5’ biotin-GTCCACATTCCACCGAAG 3’) was incubated with different concentrations of TEAD3 protein in reaction buffer containing 50 mM HEPES-NaOH pH 7.0, 0.1% BSA at room temperature for 1 hour, Anti-FLAG antibody was used to detect reaction products.

Storage
