Background

Recombinant human PHF2 (PHD finger protein 2), also known by its gene symbol PHF2 and alternative names including JHDM1E (Jumonji C domain-containing histone demethylase 1E), KDM7C (lysine-specific demethylase 7C), CENP-35 (centromere protein 35), and GRC5, is a flavin adenine dinucleotide (FAD)-binding protein that functions as a histone demethylase specifically targeting dimethylated Lys-9 of histone H3 (H3K9me2) to promote transcription activation of target genes. This recombinant form, typically produced in expression systems such as E. coli, retains the key biochemical properties of the native protein, including its JmjC domain that confers demethylase activity and its PHDtype zinc finger that mediates binding to methylated histones. PHF2 plays critical roles in diverse biological processes including adipocyte differentiation where it promotes expression of major transcription factors, liver development as a coactivator of HNF4A, and activation of toll-like receptor 4-target inflammatory genes in macrophages.
